ABSTRACT

Objective To detect the expression of BMI-1, and to discuss its role in the development of gastric cancer. Methods The expression of BMI-1 protein and mRNA in gastric cancer and adjacent tissues was detected by immunohistochemical P-V method (78 cases) and semi-quantitative RT-PCR (30 cases). Results The positive expression rate of BMI-1 protein in gastric cancer patients was 66.7 % (52/78), and it was 19.2 % (15/78) in adjacent tissues, the difference was statistically significant (χ2= 35.815, P= 0.000). The expression of BMI-1 mRNA in 30 gastric cancer tissues was 0.23 ± 0.12, it was 0.03 ± 0.12 in adjacent tissues, the difference was statistically significant (t=8.372, P=0.000). The expression of BMI-1 protein and mRNA was correlated with the degree of tumor differentiation, depth of invasion, lymph node metastasis and TNM staging (all P< 0.05). Conclusion The expression of BMI-1 is closely related to the occurrence and development of gastric cancer, which may provide guidance for molecular pathogenesis, targeted therapy and prognosis of gastric cancer.

ABSTRACT

Objective To observe and analyze the short-term clinical effect of Zhenwu decoction combined with taxol and carboplatin (TC) in treatment of post-operative patients with ovarian cancer.Methods The clinical data of eighty-six post-operative patients with ovarian cancer from June 2014 to June 2015 in our hospital were retrospective analyzed, and randomly divided into control group and treatment group, 43 cases in each group.The control group received TC treatment and treatment group received Zhenwu decoction on the basis of control group with two courses, 21d of one course. The clinical effect were compared between two groups post-treatment.Results After treatment, the total efficacy in treatment group was higher than that in control group (88.37%vs.65.11%;χ2 =6.515,P0.05), the reducing rate of life quality was lower (4.65%vs.41.86%;χ2 =10.030,P<0.05), the effect of Karnofsky performance score (KPS) was higher (93.02% vs.72.09%;χ2 =6.541, P<0.05), the complication rate was lower (4.65% vs.18.60%;χ2 =4.074,P <0.05).Conclusion The clinical effect of Zhenwu decoction combined with taxol and carboplatin (TC) in treatment of post-operative patients with ovarian cancer was satisfying, which could improve prognosis and improve survival rate.

ABSTRACT

BACKGROUND:It is a key to choose an appropriate method to trans-differentiated bone marrow mesenchymal stem cells (BMSCs)into neuron-like cells for clinical therapy of neural system injury.OBJECTIVE:To observe the feasibility of the differentiation of rat BMSCs supplemented with rat dentate gyrus extract(DGE)into neuron-like cells.METHODS:DGE was applied to induce rat BMSC trans-differentiation,using 20,40,60,80 mg/L protein concentration.Neuron differentiation was measured using Western blot to screen an optimal dosage.After trans-differentiation,different concentration treated cells were collected.Morphology change was observed following differentiation under an inverted microscope.Neuron specific enolase(NSE)and NeuN expression was determined using Western blot.NeuN expression in cells was detected using immunofluorescence technique.RESULTS AND CONCLUSION:After 7 days,60 mg/L DGE was the optimal induction dose detected by Western blot.With increased concentration,NSE and NeuN expression was increased.At 80 mg/L mass concentration,NSE and NeuN expression was reduced.At 60 mg/L DGE,BMSCs following induction became long,with synapses,Immunofluorescence NeuN staining found that neuron-like cells were positive for NeuN following induction.Results indicated that DGE is an effective biological inductor that can induce BMSC differentiation.

ABSTRACT

Objective To study the significance of Helicobacter pylori (Hp) infection and the expression of caudal-related homeobox transcription factor 2 (CDX2),mucin 2 (MUC2) proteins in gastric cancer,and elucidate the prognostic significance of CDX2,MUC2 proteins in gastric cancer.Methods Ninety cases of gastric cancer were available for the study.Hp was studied by Warthin-Starry bacterium stain and CDX2 and MUC2 proteins were studied by immunohistochemical stain.Results There was no Hp infection and no expression of CDX2 and MUC2 proteins in normal gastric mucosa.The positive rate in the cases of intestinal-type gastric cancer was 67.86%(38/56) for Hp,75.00% (42/56) for CDX2,and 66.07%(37/56) for MUC2,respectively,and it was higher than that in the cases of diffuse-type gastric cancer,which was 35.29%(12/34),41.18%(14/34) and 38.24%(13/34) respectively (P<0.01 or < 0.05).In intestinaltype gastric cancer,the positive rate of CDX2 and MUC2 proteins was also higher in Hp positive cases than that in Hp negative cases,and it was the same as CDX2 protein in diffuse-type gastric cancer.Higher positive rates of CDX2 and MUC2 proteins were observed in the cases of early gastric cancer than those in the cases of advanced gastric cancer (P < 0.01 or < 0.05),also it was observed in the cases of no lymph node metastasis than in the cases of lymph node metastasis (P < 0.01 or < 0.05).Moreover,the positive rate of CDX2 protein in the cases of stage Ⅰ~Ⅱwas higher than that in the cases of stage Ⅲ~Ⅳ (P < 0.05).Conclusions Hp infection and the expression of CDX2 and MUC2 proteins are related with gastric cancer,and has close relationship with intestinal-type gastric cancer.The expressions of CDX2 and MUC2 proteins may inhibit the infiltration and metastasis of gastric cancer.

ABSTRACT

Objective To investigate the relationship between the expression of p53, ki-67 proteins and clinicopathology of human thyroid carcinoma. Methods Using SP immunohistochemical methods, the expression of p53 and ki-67 proteins was detected in 55 cases of thyroid carcinoma. Result The positive rate of p53 protein expression was 52 7%(29/55) and ki-67 proliferative index was(11 07?3 84)% in the thyroid carcinoma. The positive rate of p53 protein expression and ki-67 proliferative index increased with the decrease of the pathological grade, and the increase of lymph node metastasis and capsule invasion. Conclusion The abnormal expression of p53 and ki-67 proteins might play an important role in the pathogenesis and development of thyroid carcinoma. It has an important clinical value for early diagnosis, prognosis evaluation and proper treatment of thyroid carcinoma to determine the expression of p53 and ki-67 proteins simultaneously.
